Challenge of the day...

I have 345 hours in Call of Duty: Modern Warfare 2 Multiplayer on Steam. Haven't played that game in years though...
 
DOTA 2 - 250 hours on Steam, but that is nothing. I've probably spent over 1000 hours playing Heroes III and Galactic Civilizations II.

Honorable mention: Archlord (2 years of top level PvP, best guild in the game, I was a guild officer for some time - so I spent a lot of hours there too: definitely more than 100, though less than 1000).
